I had a Mark III (just sold it this week) and now have a Studio Pre with Mesa 50/50. The cab I have been runnning through is a custom made 2x12 cab with one Weber Blue Dog and one Weber Silver Bell. My tone sounds great but I feel as if these speaker are not giving me as heavy a crunch sound and chug in the bottom end as I would like. My question is, can speakers make a difference on perceived gain? Would say a V30 be a better speaker for heavy music over the Webers?

most definitely. there are speakers out there tuned to project certain frequencies for each style of music. for example, I wouldn't use a tone tubby to do heavy metal music.

go to celestion.com and search around a little...they show you descriptions of different speakers for different genres of music.

For extreme crunch on a Mark series Mesa, I'd take an EV12L or a Vintage30
