I have a US Single Recto that I run on 240v in Australia through a step down transformer. Twice now I have mistakenly plugged it sraight into 240V. The first time I blew the fuse but it was all good. This time I think I have damaged it. The clean channel is fine but the O/D channel has lost significant out put as well as definition. I guess once I get it fixed I should look at replacing the power tranny with a 240v huh.

If the clean channel is fine, you did not blow the PT.

Loss of definition and volume on the dirty channel only does sound like a V3 issue, and this would be a definite possibility given what would happen if the amp was over-voltaged for a short while before the fuse blew - the tube filaments would run far too hot which may damage the filament insulation, and the usual cause of V3 failure is a cathode-to-filament short.
