Hi,
I'm trying to find the best (to me) preamp tube for my DC-5.
This amp has 6 12ax7 but I know that the V5 is for reverb, and V6 for phase inverter.
Will the others 12ax7 (V1, V2, V3, V4) affect the tone?
I have the schematic here but I can´t understand it.

The other positions will affect tone, V1 having the most impact. I don't own a DC-5 so I don't know the details, but I'm assuming that each pre-amp tube handles the tone for each channel...but this isn't a four channel amp.... are you sure there are 6 pre-amp tubes? not 5?
 
Yes, six 12ax7 on a two channel amp.

The schematic is like this:

Lead Channel
V1a-V2a-GAIN-V2b-V3a-TONE CONTROL

Rhythm Channel
V1b - TONE CONTROL - GAIN - V4b -V4a
...........................................................................................................
V5 on Reverb

V5a-REVERB-V5b
...........................................................................................................

V3b on Send Return:

FX RETURN - V3b - OUTPUT LEVEL
...........................................................................................................

And V6 on Phase Inverter
...........................................................................................................

So, will all the first four preamp tubes affect the tone?

V1,2,3,4 will all affect tone. So will the 6L6s.
