I bought a new set of preamp tubes (Mesa), when I got home
and opened them up, I noticed that one of them was different.
One of them says Russia on it and the inside looks a little different.
Is this a good thing or a bad thing? should I put it in the V1
slot or another slot?

The Russian labled tube is now a "New Old Stock" mesa tube. Their current tube line are relabeled JJ's which are said to be of Slovakian origin. The Russian tube has a bit more gain than the new JJ's and to my ear a little brighter. The V1 slot is a good place to start, but experiment, depending on which amp you have you may like it else where.
