As with all amps V1 and V2 are the root of the tone. My question is "does the Mark IIb's lead drive tube in V3 and the Cascaded lead drive tube in V4 effect the overall tone, or does it just overdrive the tone already generated in V1 and V2?"

All opinions/suggestions thoughts and ideas welcomed.

Of course. The more gain you use, the more tone you actually lose. The lead channels effect has been altered in the cathodes on the lead channel to compensate for certain " Desireable" frequencies, but the higher the gain, the less control the tone stack has over it. You may actually notice less effect of your tone controls when using the highest gain settings.
