I'm planning to pick up a Mark V, but I have some questions on how the Loop Assign function works.

It's my understanding that if I set the Loop Assign to Channel 3, the loop will be on by default when I switch to Ch3, and off for Ch1 & Ch2.

Will I still be able to switch the loop on and off for Ch1 & Ch2, or is the loop now permanently wed to Ch3?

Similarly, once I'm in Ch3, will I be able to switch the loop off?
 
I'm pretty sure the footswitch only works on the loop if you have the loop-assign mode in the back on footswitch. If you have it set to channel 3, it will always be on on channel 3, and never on on the other channels, even if you stomp the footswitch.
 
If you have the loop set to "on" for channel 3, it is always on. Channels 1 and 2 will be whatever you left it on when you were last on that channel via the footswitch, but works universally between 1 & 2 in that instance... so if you turn it on for Ch. 1 and then go to Ch. 2 it will be on. If you have it off for Ch. 1 and go to Ch. 2 it will be off as well. And then when you go to Ch. 3 it will be on per your setting.
 
Can the loop be switched off via the footswitch when Ch3 is engaged?
 
S-daisy is correct. The question you need to ask yourself is, "which is better for you; setting the loop to be on when you switch over to that particular channel or have the flexibility to turn on and off in all channels?"

The way I have mine set is always on for Ch. 1 and toggle on/off for Ch. 2 and Ch. 3. I have a delay with a bit of modulation on the repeats for a nice shimmery effect, but not overly chorus'ed. It works well in all 3 channels, but always on for Ch. 1. It's just short of Midi for me. I actually have a midi setup with an Amp Gizmo and Mastermind Midi footswitch, but for what my needs are, I actually get more flexibility this way since I need less equipment and my F/X needs are minimal. The Mark V is really set up well for this method.
