One of my power tubes broke, so I removed the matching pair for the time being. Does this affect my tone since I believe it's only running 50 watts?
 
it may because you are achieving powertube saturation at a lower volume. its not gonna be like "whoa somethings way different" more like, "it sounds like its cranked but its not as loud"
 
As kylek said, it won't be drastic but I imagine you'll be able to tell. It enables you to acheive power tube distortion at lower volumes, aside from that it won't be a gamebreaker for you.
 
Did you remove the associated rectifier as well?
