To change to speaker load, should I power down, or can I just go to standby?

If I wanted to change from 2/2x12 cabs, to 1/2x12 cab?

Will only going to "standby" potentially damage to amp from the sudden change in loading?
 
/agree power down completely..Only a few minutes difference and could save a ton in the end.
 
Yes, absolutely power down. Standby puts your tubes on heaters but otherwise your amp is up and running and should have a load on it. Your transformer will thank you.
