V1 is the 12AX7 closest to the input jack. In your photo, that would be the tube with the shield closest to the transformer.
V2 is to immediately to the right of V1.
And so on.
V5 is the phase inverter, and is between the row of 12AX7s (V1 thru V4), and the power tubes.
The power tubes are arranged in pairs like this: A-B-B-A.
The outside pair (A-A) is the Simul-Class pair if you have this option.
You may use 6L6s or EL-34s in these positions ONLY.
The inner pair (B-B), is ALWAYS 6L6s.
Here's the manual download page from the Mesa website:

The manual explains which tube does what.
Hope this helps :D

Isn't the tube with the shield V1? If you look at the front of the chassis that tube is the one closest to the input jack and is covered by the metal shield.

Taken from the Mark III manual:
The V1 socket is the one located closest to the Input jack or, as seen from the rear, it is the socket furthest to the right in the
front row. The V1 tube is the only one covered by a silver metal shield can. (Shields are not needed for the other tubes and may
actually make them more susceptible to noise.)

Which would make you labeling in reverse. V1 is critical for tone changes.

Also hopefully someone else can jump in. It looks like V4 has been removed (is this standard on the R2 mod?)
My Mark 3 Green Stripe is exactly the same way. And I believe this is the Reverb tube.
