The answer is different based on the specific amp model.
On the ones where the answer is YES (tank must be plugged in),
not doing so can harm a transformer.
I'd check direct with Mesa to be 100% sure.
I also did this on a Red III Short Head. I wanted to keep both the reverb and presence active so I put
the 250K audio pot where the DI out Volume was.
Really don't understand anyone (yet alone Mike B.) saying it couldn't be done.

I'd also like to have a gain control on R2 along with the volume mod.

When in R2 pull the shift on the bass control. The added
fullness in the low mids seems to smooth out the top end,
which gives the allusion of...
I have a red stripe also and it doesn't like my Strymon stuff in the loop.
The overall effect and tone just seems off. An MXR carbon works great though.
